It would be advisable to check the CO2 level of the incubator you are using. Or, if you do not have an incubator fed with a CO2 supply and have not done so already - gas the flask with CO2. The recommended level of CO2 for COS-7 cells is 5%. The pinker than usual appearance of the medium may be caused by too low a level of CO2. Commonly cell culture media contains phenol red which responds to changes in pH. The colour of the medium gives an indication of the pH – from purple/pink at alkaline pH through to yellow when acidic. The concentration of CO2 in the environment and that produced by the cells as they proliferate, in addition to lactic acid production, affects the pH of the medium. The higher the concentration of CO2 the more acidic the medium becomes. If the medium is purple/pink it suggests it is alkaline which could be due to too low a level of CO2 in the gaseous phase.
