Human Genomic DNA Production

The sequencing of the human genome and the recent advances in the field of human genomics has enabled the identification of new polymorphisms that are associated with particular genetic susceptibilities. ECACC’s extensive collections of over 40,000 cell lines provides a prime resource for this growing research field and has allowed ECACC to diversify into new product areas. Our scientists have developed methods for extracting DNA from its collection of cell lines. The standardized DNA is authenticated using STR PCR and is ready to use in convenient cost effective formats.

Facilities
Two well equipped, dedicated laboratories are used and one is set up for the production of large quantities of genomic DNA from any of ECACC's cell lines. The scale-up laboratory is equipped with class 2 microbiological safety cabinets that operate to containment level 2. ECACC has the capacity to use hundreds of roller bottles during the scale-up process.

Capability
In the year 2002, ECACC manufactured over 1,500,000 micrograms of DNA from over 2,000 different sources. ECACC has a capacity  to perform more than 500 large-scale DNA extractions per month. Although DNA is extracted primarily from human cell lines, DNA can be extracted from any of ECACC's celll lines. State of the art barcode driven tracking and inventory systems, in addition to cell line and final product authentication, ensure confidence in product identity.   • Human Random Control (HRC) DNA
  • Ethnic control DNA
  • Primate DNA
  • Genetic disease Human DNA
  • Human Random Control DNA extracted from leukocyte reduction filters

    Quality control
    To ensure the highest quality of genomic DNA, the extracted DNA is subjected to a number of quality control procedures to determine the molecular weight, integrity, purity, PCR performance and long term stability.
